PMA is often used in combination with Ionomycin to stimulate immune cell activation and is used to induce the production of cytokines in in vitro cell cultures. Often used in combination with protein transport inhibitor Kit-1 (Cat:IK-PTI-1). At the same time, the protein transport inhibitor Monensin or Brefeldin A can also be added to inhibit the exocytosis of cytokines to accurately analyze the expression levels of cytokines. If performing immunoassays for secreted Proteins (e.g. ELISA, Western blot, or multiplex protein assays), there is no need to treat cells with Monensin or Brefeldin A.
